Targeting Leukemia Stem Cells to Expand Treatment-Free Remission in CML
This narrative review maps emerging leukemia stem cell surface markers, signalling pathways, and epigenetic regulators that may extend treatment-free remission beyond what tyrosine kinase inhibitors alone can deliver.

Novel therapies for CML aim to overcome resistance and improve remission.
Improving management of chronic myeloid leukemia (CML) by investigating novel therapeutic targets could increase treatment-free remission (TFR) rates. New molecular and immunotherapeutic targets have been identified that focus on leukemia stem cells (LSCs) and their role in treatment resistance. Current tyrosine kinase inhibitors (TKIs) are effective but limited by resistance; exploring these new targets can lead to better outcomes and potential curative approaches. Emerging targets in LSCs for CML therapy could enhance TFR rates and possibly achieve curative outcomes.
